Growth of  Monostroma  sporophytes in different conditions Felix bast
Objectives What are the optimum values for salinity, temperature and media concentration for the sporophyte growth? Which media is better, West and Mc Brides modified ES or Porfiran? Will frequent transfers (daily) lead to increased growth? Pasteurization better than autoclaving? Surface Sea Water or Deep Sea Water, best for growth?
Methodology Sporophytes from the samples originally isolated on the same week (03-2006) were grown in different conditions. Transferred to fresh media in every 4 days except in the control (daily transfers) Photomicrographs were taken in every 4 days. Group of easily identifiable (same pattern or location) cells were selected and 4X print-outs were made in order to photograph same group of cells each time

Methodology Using the public domain software: ImageJ 速 , area of at least 3 cells were measured and mean value is recorded

Results Half the normal salinity value is found to increase the growth rate. 15 属 C is best for the growth of sporophytes, then 25 属  C and 20 属 C Daily transfers to fresh media found clearly to increase the growth rate. Deep Seawater observed to be better than Surface Seawater for the growth Pasteurization of seawater is observed to be having slight advantage than autoclaving. Porfiran is found to be better than WMB-ES for the growth of sporophytes
Prospective research Same experiment to be continued for one month (till 12-8) Detailed life-history studies of asexual and sexual strains with detailed micropotographs Optimization of synchronous zoospore release in  Monostroma
