Matrix-M is an adjuvant formulated from saponins extracted from Quillaja saponaria Molina. This study examined the early inflammatory response to Matrix-M in pigs. In vivo and in vitro experiments showed Matrix-M did not cause harmful inflammatory reactions in piglets. Histological examination found leukocyte infiltration and tissue damage at the injection site in older pigs. Transcriptional analysis revealed Matrix-M activated genes related to myeloid cells and plasmacytoid dendritic cells. Genes for cytokines, chemokines and their receptors were upregulated at the injection site and draining lymph node.
