Chromatiscope is a lab-in-a-smartphone microscope, fluorometer, spectrometer, and colorimeter that allows students to explore the microscopic world and analyze samples. Nanolive’s 3D Cell Explorer allows for the creation of very powerful 3D images and 4D time lapses of living cells with very high spatio-temporal resolution (x,y:180nm; z:400nm; t:1.7sec). Moreover, imaging with the 3D Cell Explorer does not require the use of any labels since the microscope directly measures the refractive index of the different substructures of the cell. However, in the context of molecular or cellular biology investigations, it can be useful to follow fluorescent markers combined with the refractive index distribution to validate specific structures or to correlate fluorescent signals and cellular states. For this purpose, Nanolive developed the 3D Cell Explorer-fluo (https://nanolive.ch/fluo/) – a complete solution that combines high precision tomographic data with high quality tri- or fourchannel epifluorescence provided by a CoolLed module (https://www.coolled.com/). In this application note we will present the time lapse imaging of mouse embryonic stem cells (mESCs) that have been genetically modified to express the fluorescence ubiquitination cell cycle indicator (FUCCI), a two-color (red and green) indicator that allows to monitor the cell cycle phases. We will explain how to use the 3D Cell Explorer-fluo to record movies that require both fluorescence and 3D refractive index imaging and will propose a solution to analyze the resulting time lapse experiment.

Drug discovery and development is a long and expensive process over time has notoriously bucked Moore's law that it now has its own law called Eroom's Law named after it (the opposite of Moore). It is estimated that the attrition rate of drug candidates is up to 96% and the average cost to develop a new drug has reached almost $2.5 billion in recent years. One of the major causes for the high attrition rate is drug safety, which accounts for 30% of drug failures. Even if a drug is approved in market, it could be withdrawn due to safety problems. Therefore, evaluating drug safety extensively as early as possible becomes all the more important to accelerate drug discovery and development. This talk provides a high-level overview of the current process of rational drug design that has been in place for many decades and covers some of the major areas where the application of AI, Deep learning and ML based techniques have had the most gains. Specifically, this talk covers a variety of drug safety related AI and ML based techniques currently in use which can generally divided into 3 main categories: 1. Classification 2. Regression 3. Read-across. The talk will also cover how by using a hierarchical classification methodology you can simplify the problem of assessing toxicity of any given chemical compound. We will also address recent progress of predictive models and techniques built for various toxicities. It will also cover some publicly available databases, tools and platforms available to easily leverage them. We will also compare and contrast various modeling techniques including deep learning techniques and their accuracy using recent research. Finally, the talk will also address some of the remaining challenges and limitations yet to be addressed in the area of drug safety assessment.

It is estimated that there is in excess of 150 million tons of plastic materials in the world’s oceans. Much of this pollution consists of large items such as discarded drink bottles and plastic bags. However, there is increasing research into the amount of much smaller materials, termed microplastics, in the river and ocean systems which present a different type of problem for marine life. Many cosmetic products, such as facial scrubs, toothpastes, and shower gels, currently contain microplastic beads as abrasive materials. These microplastics, which are typically submillimetre in size, get washed down the sink and are too small to be filtered by sewage treatment plants consequently ending up in the river systems and ultimately in the oceans. These microplastics can be ingested by marine organisms and fish and end up in the human food chain. In 2014 a number of U.S. states banned the use of microplastics in cosmetic formulations and most cosmetic companies are voluntarily phasing out their use. Infrared (IR) spectroscopy is the established technique for identifying polymer materials and has been used extensively for identifying large (over 100 micrometer) polymer materials. The Spectrum Two™ is a portable FT-IR spectrometer that can operate from a battery pack and has been used on boats for immediate identification of these polymers.1 For microplastics, down to a few micrometers in size, an IR microscope can be used for the detection and identification of these materials.
