corticosteroids and antiemetics in rat plasma by solid phase extraction and ultra-
performance liquid chromatography-tandem mass spectrometry: Application to
pharmacokinetic interaction studies
This work deals with the development and validation of a sensitive and selective ultra-
performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS)
method for the simultaneous determination of selected tyrosine kinase inhibitors
(TKIs); gefitinib (GEF), erlotinib (ERL), along with corticosteroids (dexamethasone
DEX, prednisolone PRED), and the antiemetic ondansetron (OND) in rat plasma
samples. Steroids and antiemetics are usually used with GEF/ERL to improve
treatment- and disease-related symptoms in patients with cancer. They may affect the
cytochrome 3A4 (CYP3A4)-mediated metabolism of the co-administered TKI. Thus,
this study is useful to study the possibility of drug-induced interactions that could be
encountered following their co-administration. This study can be readily applied in
therapeutic drug monitoring (TDM) in patients receiving these drug combinations as
well as investigation of possible drug interactions between the cited TKIs and
DEX/PRED/OND.
