- 1. Reports to The post holder will report to Professor Mike Ehrenstein Context UCL is one of the top Universities in the world with superb facilities for biomedical sciences and its translation to human disease. The successful candidate will join Professor Ehrenstein’s group which focusses on translational research in autoimmune rheumatic disease spanning disease based mechanistic science to early phase clinical trials. Work within Professor Ehrenstein’s team aims to translate advances in the understanding of autoimmune rheumatic diseases into improved diagnosis and therapy. Its close link to UCLH provides an excellent interface between science and medicine. Professor Ehrenstein strengthens these links by being actively involved in the recruitment of patients for studies from rheumatology out-patient clinics. The work will be carried out at the Centre for Rheumatology in the Rayne Building, which has outstanding flow-cytometry infrastructure and technical support. This project is in collaboration with Dr Muhammad Shipa at UCL, who has a wealth of knowledge on computational analysis, and Dr Louisa James at QMUL, and some research activity may be based at QMUL. The post is funded by Versus Arthritis and is available for 1 year in the first instance. Main purpose of the job The research objective is to apply molecular and cellular techniques to understand the role of B cells and identify biomarkers of response to B cell targeted therapy in patients with systemic lupus erythematosus (SLE). The project is supported by a research nurse who collects samples from patients. Bioinformatic support will also be available. The research will be laying the foundation for a biomarker driven clinical trial and provide mechanistic understanding of response to B cell targeted therapy. Duties and responsibilities To receive, process and store biological samples from patients and healthy invididuals as a comparator. Some samples will be obtained from industry and academic partners. To analyse autoantibodies, cytokines and B cell subsets in patients with SLE using a range of approaches including multiparameter Flow Cytometry, ELISA, RT-PCR and RNA sequencing as appropriate. The need for flexible working hours may be required to analyse and purify fresh rather than frozen samples. To liaise with bioinformaticians to process and analyse RNA sequencing data. Careful documentation of sample arrival, followed by processing and storage is required. Robust audit of sample movements. Careful documentation of results and analysis including generating protocols and SOPs. Forward planning of equipment and material needs. Maintenance and ordering of laboratory stocks of chemicals, media and other consumables required for the project. Ensuring correct storage and handling of biochemicals and biological materials. Assistance in the maintenance of the laboratory safety information database. Responsible for the safe disposal and storage of hazardous waste in accordance with local rules and legal requirements. Adherence to relevant safety rules, including assessment of risks, using relevant rules. To keep abreast of the literature in the fields of research relevant to the post holder’s project. - 2. To prepare and present findings of research activity to colleagues for review purposes. To contribute to the drafting and submitting of papers and reviews to appropriate peer reviewed journals. To prepare progress reports on research for funding bodies as required. To contribute to the preparation and drafting of research bids and proposals. - 3. Person Specification Criteria Essential or Desirable Qualifications, experience and knowledge PhD or equivalent in Biochemistry/Immunology/Biomedical Sciences. Appointment at Grade 7 is dependent upon having been awarded a PhD; if this is not the case, initial appointment will be at research assistant Grade 6B with payment at Grade 7 being backdated to the date of final submission of the PhD thesis. E Prior experience of multiparameter flow cytometry as applied to immunology E Experience with sterile cell culture technique and cellular immunology E Handling and processing of clinical specimens, including serum and PBMC isolation and cryopreservation. E Good knowledge of statistics E Evidence of high quality publications of original research E Basic computational analysis of complex datasets (bioinformatics) D Molecular biology skills including experience of RNA sequencing D Skills and abilities Ability to perform and analyse multiparameter flow cytometry E Skills in molecular and cellular immunology including sterile culture techniqes E Experience with RNA sequencing D Basic computational skills and statistical analysis E Ability to work effectively within a team environment & on own initiative, promote teamwork and motivate colleagues E Excellent communication skills/verbal and written.
