Electrical stimulation uses small electrical currents to stimulate nerves and muscles. It can be used to reduce pain, prevent muscle atrophy, and improve strength. In neurological rehabilitation, it is used for spasticity reduction, edema control, and functional electrical stimulation to improve gait. The Empi Continuum device has two channels and customizable settings to provide timed electrical stimulation sessions to targeted areas like the shoulder, wrist, quadriceps, and plantar flexors. Electrical stimulation must be applied carefully based on a person's needs and medical conditions.
Spasticity is a motor disorder characterized by velocity-dependent increase in muscle tone and exaggerated reflexes resulting from hyper-excitability of the stretch reflexes as part of the upper motor neuron syndrome. It is caused by interruption of descending inhibitory pathways and alteration of central inhibitory commands leading to rearrangement of spinal cord circuitry and peripheral changes in the muscle over time. Spasticity is not a single symptom but part of a movement disorder defined by features like hypertonia, abnormal reflexes, dystonia, resistance to movement, and abnormal posturing affecting coordination and mobility. A multifaceted approach is needed for management including prevention, physiotherapy, pharmacological and surgical interventions to address both neural and non-neural factors.
This document describes constructing deletion mutants of several genes in the uropathogenic E. coli strain UTI89. The key steps are:
1. PCR was used to generate gene disruption cassettes for spr, prc, eco293-sinI, and fimS.
2. The cassettes were electroporated into UTI89 cells expressing Red recombinase and plated on chloramphenicol to select mutants.
3. PCR confirmed successful disruption of the spr gene, replacing it with the cassette.
4. The cassette was then evicted by expressing FLP recombinase from pCP20, leaving only the gene deletion.
